More about the position
The main objective of the position is to analyse T cell immune responses in blood samples collected from participants of a clinical trial at the center. This clinical trial is a proof-of-concept study to target the metabolism of activated gluten specific T-cells in celiac disease. The candidate will be in charge of collection and analysis of blood samples collected as part of the trial. Specifically, this will involve isolation of gluten specific T-cells and phenotyping by multicolor flow cytometry, single cell isolation and sequencing of T cell receptors by next generation sequencing. The project is focused on the concept of targeting gluten specific CD4+ T cells as a potential cure for celiac disease. Research from the center has demonstrated that gluten specific CD4+ T-cells have a very narrow phenotype and a metabolic program that together can be exploited to specifically target and remove these cells.
The center has initiated a clinical trial (NCT04806737) where participants with well treated celiac disease will be subjected to a short-term gluten challenge together with placebo or the drug Teriflunomide, which inhibits oxidative phosphorylation in recently activated T cells. The project of the candidate is to monitor the gluten specific T-cell response in blood by flow cytometry and T cell receptor sequencing. The candidate is therefore required to have extensive experience from work with human T-cells isolated from blood and strong laboratory skills in methodology relevant for the projects (see “Qualification requirements” for further details.)
A main goal of the K.G. Jebsen Coeliac Disease Research Centre is to translate advances in the basic understanding of the pathogenesis of coeliac disease to the clinic – to improve disease diagnostics, to implement novel treatments and to identify new therapeutic targets for this common disease. The work of this project explores a ground-braking and novel approach to cure coeliac disease.
The successful applicant will join a 20-member group within an interactive multidisciplinary centre of scientists, in a strong academic environment, focusing on immune dysregulation in autoimmune disease (see http://www.med.uio.no/klinmed/english/research/centres/kgj-coeliac-disease/index.html and http://www.med.uio.no/cir/english/research/groups/sollid/ for further information).
The main purpose of the fellowship is to qualify researchers for work in higher academic positions within their disciplines.
Qualification requirements
We are looking for a highly motivated and structured candidate with excellent laboratory skills and good collaborative qualities.
For appointment, the applicant must have a project-relevant PhD degree. Doctoral dissertation must be submitted for evaluation by the closing date. Appointment is dependent on the public defence of the doctoral thesis being approved. Please also refer to the regulations pertaining to the conditions of employment for Postdoctoral research Fellows.
Fluent oral and written communication skills in English.
The following qualifications are required for the position and will count in the assessment of the applicants:
wet-lab experience with human T-cells and multiparameter flow cytometry
wet-lab experience with single cell and bulk TCR sequencing from cells isolated from blood
experience with analysis of immune cell receptor sequencing data
strong organizational and project management skills
a strong publication list
research background in human immunology and autoimmune, gastrointestinal disease is advantageous.
