The Department of Psychiatry and Neurochemistry is located at the Institute of Neuroscience and Physiology at the Faculty of Medicine at Sahlgrenska Academy, University of Gothenburg. At the Department research is conducted on human behaviour, neuropsychiatric processes as well as psychiatric treatment and diagnosis in close collaboration with Västra Götaland region and a number of international partners. We participate in the undergraduate programmes in Medicine and within The National Research School in Psychiatry/Southwest Sweden. Duties   

The PhD education in Medical Science comprises carrying out a scientific project and completing at least 30 credits of courses at third-cycle level. Project: To facilitate the accurate clinical diagnosis and the development of effective treatments for neurodegenerative disorders, in vivo biomarkers which reflect the underlying pathology and subsequent neurodegeneration are urgently required. In the department of Psychiatry and Neurochemistry at the Sahlgrenska Academy, this Ph.D project will aim to develop targeted methods of novel marker for astrocyte pathology in cerebrospinal fluid and the discovery of novel biomarkers reflective of astrocyte pathology in blood using broad range of proteomic methods. The prospective PhD student will gain experience in both biochemical techniques, molecular imaging methodologies and bioinformatics. 

Education at third-cycle level comprises four years of full-time study, and leads to a doctoral degree.  As part of your employment as a doctoral student, you may have departmental duties corresponding to up to 20 % of full-time employment, distributed throughout your study period. Departmental duties usually consist of teaching at first- and second-cycle levels, but may also include research and administration.    

Eligibility   

Education at third-cycle level requires general eligibility and specific eligibility as set out in the general syllabus for the subject.  

The general eligibility requirements for education at third-cycle level are:    
1. having completed a degree at second-cycle level, or
2. the fulfilment of course requirements totalling at least 240 credits, of which at least 60 credits must be at second-cycle level, or

Specific entry requirements for this subject, according to the general syllabus, are:

Assessment criteria   

The selection of applicants who are considered to have the ability required to benefit from the study programme and who meet the general and specific eligibility requirements are assessed based on the following documented qualifications:
- specific knowledge and skills within the subject area and related research fields
- experience of scientific theory and relevant research methodology
- scientific analysis and presentation verified through thesis, degree project, scientific journals or the like.    

Documented laboratory experience is meritorious. Experience of biochemical method, Astrocyte biology and the understanding of coding language is desired.

Employment   

Once you have been admitted for education at third-cycle level, you will be employed as a doctoral student at the University of Gothenburg. The provisions for employment as a doctoral student can be found in ordinance SFS 1993:100. Initial employment as a doctoral student may apply for a maximum of one year, and may be renewed by a maximum of two years at a time. Project: The overall aim of our research is to determine the causal mechanisms behind severe mental illnesses, including the affective disorders major depressive disorder (MDD) and bipolar disorder (BD). In this project, we propose to identify novel predictors of treatment response and in the long run to improve outcomes for patients suffering from these diseases by enabling tailor made pharmacotherapy. By taking advantage of preexisting unique cohorts of MDD and BD patients, we will create cellular models for individuals using induced pluripotent stem (IPS) cells from meticulously characterized patients. So far, we have established cultures of several lines from BD patients and controls that are used to study neural development. We hypothesize that we can identify novel predictors of treatment response in mood disorders by using patient derived cell models. These predictors may be common to several lines of treatment, thereby revealing new knowledge on underlying pathophysiology.

Doctoral position Project-funded PhD position in Natural Language Processing    

The department of Swedish, Multilingualism, Language Technology announces a PhD position within the research environment project Grandma Karl is 27 years old: Automatic pseudonymization of research data (funded by the Swedish Research Council, https://spraakbanken.gu.se/en/projects/mormor-karl).

The research project

The position is funded mainly by a research environment grant from the Swedish Research Council on automatic pseudonymization of research data). The project leader is Elena Volodina.

Accessibility of research data is critical for advances in many research fields, but textual data often cannot be shared due to the presence of personal and sensitive information, e.g names, political opinions. GDPR suggests pseudonymization as a solution, but we need to learn more about it before adopting it for manipulation of research data.

The Grandma Karl research environment targets several aspects of pseudonymization, aiming to advance Sweden's work on open access to research data. The primary focus of this position will be  development of algorithms to automatically detect, label and pseudonymize personal identifiers in freely written texts (essays/blogs), focusing on linguistic challenges such as spelling errors, ambiguous entities, semantic constraints etc. A minor part of the research can be devoted to the analysis of type and number of personal identifiers versus acceptable protection, followed by reidentification tests to ensure that pseudonymization is effective.

The successful candidate should write their doctoral thesis within the core area of the funding project. The doctoral student is expected to cooperate with other project members (including at other partner universities), participate in project activities (e.g. meetings), participate in the departmental research related activities, as well as national and international workshops, seminars and conferences.

Project title: Longitudinal clinical screening and digital lifestyle intervention to prevent disease progression from gestational diabetes to type 2 diabetes

The Department conducts research within the area of physiology. Several projects involve a close collaboration with the Sahlgrenska University Hospital. Project: Gestational diabetes (GDM) may have serious short- and long-term consequences for both woman and child. Although glucose levels generally normalise after delivery, GDM increases the risk sevenfold for type-2-diabetes (T2D) within a ten-year period. Several studies show that the T2D risk after GDM can be reduced by lifestyle interventions and that early detection of impaired glucose tolerance is of great importance. There is currently, however, no satisfactory follow-up program after GDM leading to missed cases and delayed diagnosis of T2D. The objective of the PhD project is to measure and determine behavioural and biological factors important for disease progression from GDM to IGT/T2D, and to intervene by internet-based lifestyle modifications to reduce identified risks.

This translational project will utilize two clinical studies- the completed longitudinal PONCH study and the ongoing prospective PREPP study. In PONCH, women were screened 6-10 years after GDM pregnancies with blood sampling, clinical assessments, questionnaires and fitness testing. Metabolomics and proteomics are currently used to identify novel risk markers in this cohort. In the prospective PREPP study, we aim for earlier detection of IGT with intention to prevent disease progression. PREPP includes thorough physiological and clinical assessments 2-6 years after GDM, as well as wearable sensors monitoring behavioral risk parameters. The PREPP screening is combined with a randomized digital and tailored lifestyle intervention. As a doctoral student, you will hold clinical visits (including blood sampling, measurement of body composition and participant interviews), visits with fitness tests (including measurement of aerobic fitness and muscle strength), and measurement of physical activity in everyday life using activity monitors. You are also expected to have good oral and written proficiency in Swedish as the data collection is in Swedish.

The group focuses on the oxidation chemistry of volatile organic compounds (VOCs), and their implications for air quality and climate change. As a doctoral student, your research projects will be associated with ongoing activities within your supervisor’s research. The current research projects range from chamber studies to field campaign measurements. These projects use advanced mass spectrometric techniques (e.g. a chemical ionization mass spectrometer with filter inlet for gases and aerosols (FIGAERO-CIMS) and ancillary instrumentation for the characterization of organic compounds in the gas and particle phase in different environments. The aim is to identify the sources and to determine the key oxidation processes of organic compounds in these environments, with a specific focus on organic nitrates (ONs). Organic nitrates (ONs), are an important reservoir of nitrogen oxides (NOx) and play a crucial role for secondary organic aerosol (SOA) and tropospheric ozone (O3) production.

Duties   

The PhD position, which is partly funded by resources from FORMAS-the research council for sustainable development, will have a research focus on developing a molecular-level understanding of key processes that are of importance for the occurrence and properties of secondary organic aerosol and ONs. This project includes both fieldwork and laboratory experiments with a FIGAERO-CIMS, and chemistry modeling. Specifically, the candidate will

  • perform both ground-based and airborne observation with an unmanned aerial vehicle (UAV)-based sampling platform to detect chemicals for both urban and rural environments.
  • perform laboratory experiments to constrain physiochemical processes responsible for the SOA and ONs’ formation and evolution
  • use the obtained knowledge from field observation and lab experiments in models to better assess how ONs influence regional air quality.

The PhD education includes the development of skills and knowledge in general tools applicable to the research. E.g. one will achieve detailed knowledge within advanced mass spectroscopy and data analysis using programming software such as of Python, MatLab, or similar. For mechanistic evaluation the candidate will use kinetic modelling for use in chemical transport models. Any pre-knowledge using these tools are beneficial but not a requirement. The research topic is within atmospheric chemistry implying applicants would benefit from having a background in chemistry or alternatively within physics, geosciences, or environmental science.

Project: Alzheimer’s disease (AD) is one of the most common neurodegenerative diseases and it is estimated that more than 100,000 people are affected in Sweden. Research over the last 30 years has shown that the pathological changes in the brain can be attributed to different protein aggregates composed of amyloid-β (Aβ) and tau protein in the form of plaques and tangles, respectively. Studies have shown that in AD there is a very strong association between tau aggregates and Aβ-containing plaques. However, neither the mechanism behind tau aggregation nor the actual connection between Aβ and tau is currently known. It is not known how these processes lead to AD symptoms or what triggers the disease. Moreover, tau aggregation without Aβ pathology can occur in other neurodegenerative diseases, commonly called primary tauopathies. Biochemical tests (biomarkers) have received increasing attention, as they are valuable tools in clinical studies to better understand the progression of the disease, to monitor the effectiveness of new drugs, and to help in diagnosing the disease at an early stage in which disease-modifying drugs have optimal effects. Suitable methods to identify and measure such biomarkers are immunoassays and mass spectrometry (MS). The project aim is to reveal the pathological progression of tau protein in AD as well as other tauopathies. In particular, the project aims at finding links between the two major brain changes in AD; extracellular Aβ-plaques and intraneuronal tau tangles, and to investigate if tau accumulations associated with Aβ-plaques are different in regards to their molecular composition compared with tau accumulations formed independently.

The research group is a favourable composition for etiopathogenetic and translational research in the field of Alzheimer's and other brain diseases, with both doctors and biochemists (see https://www.gu.se/en/research/neurochemical-pathophysiology-and-diagnostics). The laboratory is well equipped, with a broad range of instruments for protein chemistry, proteomics, and immunoassays. We co-operate with several academic research groups, nationally and internationally, as well as with industrial partners

We are looking for a PhD candidate with good biochemical knowledge and experience in using immunoassays and mass spectrometry (high resolution LC-MS/MS). The work requires good interaction and communication skills and that you are careful and methodical. To be successful, the candidate will need to have a strong commitment and a positive approach, being able to work in an interdisciplinary group as well as independently towards the goals expressed in the project plans. Project
Background: Our group described one of the first cases of human infection with a previously unknown tick-borne bacterial species, N. mikurensis, 13 years ago. Our theory is that the bacteria give rise to latent infections of the vascular endothelium, which can become symptomatic when B-cell defenses decline. Chronic activation of B-cells may lead to autoimmunity and/or malignant transformation of B cells. We have recently published a study launching the hypothesis that N. mikurensis can cause malignant B-cell lymphomas.

Goals: To increase the knowledge on protective and pathologic B-cells responses evoked by N. mikurensis infection. To determine the origin and maturational grade of lymphoma B-cells from infected patients, and to assess if they are antigen-dependent, i.e., driven by bacterial or endothelial antigens. To identify bacterial antigens recognized by antibodies and the protective antibody isotypes for development of high-throughput antibody diagnostic methods for N. mikurensis. This is required to determine the prevalence of N. mikurensis in different patient and population groups, the cut-off antibody levels for when a latent infection reactivates, and the specificity of antibodies produced by lymphoma B-cells.

Project design: Mononuclear cells from patients with malignant B-cell lymphomas, with and without N. mikurensis infection, are stimulated with bacterial and endothelial antigens and phenotyped regarding 23 different B-cell markers, and the generated multiparameter data is analyzed by Helios mass cytometry and cluster analyses. Comparisons are made with the B- and T-cells responses evoked by the infection in patients with normal immune defenses and without malignant B-cell lymphoma. In silico analyses of the N. mikurensis genome, bacterial extracts, and recombinant bacterial proteins and peptides are employed to develop high-throughput antibody diagnostics for N. mikurensis. The image flow cytometry method for antibody analysis is used for comparison.

Project: Inflammatory arthritis including rheumatoid arthritis and spondyloarthritis are chronic diseases characterized by inflammation of the joints. The use of novel therapies has led to substantial improvements in clinical outcomes for patients suffering from inflammatory arthritis. However, a large unmet clinical need persists as 30-50% of patients with moderate or severe inflammatory arthritis have no- or suboptimal improvement in their symptoms despite these treatments. Thus, better understanding of disease pathogenesis is urgently needed to aid in the development of new drugs to reduce suffering of patients unresponsive to the currently available therapies.

The early phase of the disease and the flares are characterized by a heavy infiltration of neutrophils to the inflamed joints and there are increasing evidence which imply neutrophils as important players in the pathogenesis of inflammatory arthritis. Neutrophils are important regulators of inflammatory processes with the capacity to eliminate invading microbes and restore homeostasis during tissue damage. Yet, the precise mechanisms behind how neutrophils are recruited to the joints of patients with inflammatory arthritis, and how they contribute to joint damage are largely unknown. It is also largely unknown if neutrophil-derived inflammatory mediators are present and aid in driving and/or terminating the inflammatory processes in patients with inflammatory arthritis. The main goal of this PhD project is to gain mechanistic insights into the role of neutrophils in the pathogenesis of joint inflammation, and to identify alternative neutrophil-derived biomarkers that can aid physicians to better monitor and more effectively treat patients with inflammatory arthritis.

Employment   

Once you have been admitted for education at third-cycle level, you will be employed as a doctoral student at the University of Gothenburg. The provisions for employment as a doctoral student can be found in ordinance SFS 1993:100. Initial employment as a doctoral student may apply for a maximum of one year, and may be renewed by a maximum of two years at a time. A doctoral student may be employed as a doctoral student for a maximum of eight years, but the total period of employment may not be longer than the equivalent of full-time education at third-cycle level for four years.   

Location: Sahlgrenska Academy, Institute of Medicine, Department of Rheumatology and Inflammation Research. Extent: 100%. First day of employment: as agreed.

The University applies a local agreement on salaries for doctoral students.
