Doctoral student in Medical Science - Function and gene expression of single beta-cells

The University of Gothenburg tackles society’s challenges with diverse knowledge. 56 000 students and 6 600 employees make the university a large and inspiring place to work and study. Strong research and attractive study programmes attract scientists and students from around the world. With new knowledge and new perspectives, the University contributes to a better future.

Doctoral position in Medical Science

Project title: Function and gene expression of single beta-cells

The Department of Physiology is located at the Institute of Neuroscience and Physiology at the Faculty of Medicine at Sahlgrenska Academy, University of Gothenburg. The Department conducts research within the area of physiology. Several projects involve a close collaboration with the Sahlgrenska University Hospital. We offer undergraduate courses as parts of study programs in Medicine, Dentistry, Pharmacy, Speech and language pathology, Dental hygiene, Occupational therapy, Physiotherapy and the bachelor program of Cognitive Science. The research groups at the Department represent a broad range of research areas within the field of physiology.

General information about being a doctoral student at the University of Gothenburg can be found on the university's doctoral student pages: https://medarbetarportalen.gu.se/doktorand/?languageId=100001&skipSSOCheck=true

Duties

The doctoral student must also write a scientific compilation thesis or monograph corresponding to at least 120 credits. For more information about third-cycle studies at Sahlgrenska Academy, see https://www.gu.se/en/sahlgrenska-akademin/doctoral-studies

Project: Type 2 diabetes (T2D) is an escalating health problem of enormous proportions. Although existing drugs reduce hyperglycaemia, they fail to stop the gradual beta-cell failure that is the key determinant of disease progression. There is consequently an urgent need for improved knowledge of the cellular mechanisms that cause beta-cell failure. We have recently demonstrated that human pancreatic beta-cells lose their mature phenotype in T2D. Interesting reports have also shown that there are pools of beta-cells within a pancreatic islet that differ in secretory response to glucose and that the proportion of such pools is altered in T2D. This is yet a largely unexplored area, but the new findings emphasise the need to analyse the single-cell phenotype to better define the exact changes of beta-cell function and expression in T2D and enable treatment that more precisely counteracts secretory failure.

The doctoral candidate will use cutting-edge single-cell techniques to study the functional and gene expression changes that are associated with beta-cell failure. Genes that are consistently associated with beta-cell failure will be scrutinized in-depth by a range of cell-physiological techniques to pinpoint the mechanisms that fail in the secretory machinery when the gene is perturbed. By defining the phenotype and proportion of cellular pools in diabetic and non-diabetic conditions in a more precise manner than has previously been possible, the project is at the international forefront and has the potential to lead to new mechanistic understanding of beta-cell failure and potentially identify new targets to preserve beta-cell function, which is an urgent clinical need. The doctoral student will get unique training in cell-physiology, gene expression analysis and clinical medicine. The student will be part of a stimulating research environment with access to expertise in a diverse range of areas of relevance to the project.

Strong research and attractive study programmes attract scientists and students from around the world. With new knowledge and new perspectives, the University contributes to a better future.

At the Institute of Neuroscience and Physiology, at the Sahlgrenska Academy, we conduct research and education covering a wide range of knowledge areas. The institute comprises five different departments: Clinical Neuroscience, Health and Rehabilitation, Pharmacology, Physiology and Psychiatry and Neurochemistry.

The Institute of Neuroscience and Physiology, invites applications for a postdoctoral position at the Department of Psychiatry and Neurochemistry.

Subject area

Experimental Psychiatry

Subject area description

Schizophrenia is one of the most serious psychiatric diagnoses. Schizophrenia is characterized by so-called positive, negative and cognitive symptoms. Available pharmacotherapy is effective mainly for treatment of the positive symptoms (hallucinations, delusions), whereas the negative (anhedonia, inactivity, ”autism”-like symptoms) and cognitive symptoms are more difficult to treat and at the same time are those with most impact on the long-term prognosis for the patient. Alcohol use disorder (AUD) is a gigantic public health problem and afflicts also psychiatric patients, e.g. patients with schizophrenia. Treatments available for AUD, both pharmacological and psychological, have small effect sizes and simply are not good enough. The research group has recently patented two new treatment principles, one for the negative symptoms of schizophrenia and one for AUD. This preclinical project is aimed at further experimentally confirming and developing these new treatment principles. To this end we use a combination of behavioral models, in vivo microdialysis, neurochemistry and molecular biology. The studies will be conducted under supervision of Professor Bo Söderpalm and Professor Mia Ericson, at the Institute for Neuroscience and Physiology, Sahlgrenska Academy, University of Gothenburg.

Job assignments

The candidate will be responsible for the behavioral models and contribute to the in vivo microdialysis studies as well as the neurochemical and molecular biological analyses. The candidate will also be expected to compile data, draw figures and write initial versions of research articles and funding applications.

Eligibility

The eligibility criteria for employing teaching staff are set out in Chapter 4 of the Higher Education Ordinance and in the Appointment Procedure for Teaching Posts at the University of Gothenburg.

To be eligible for appointment as a postdoc, the applicant is required to have a doctoral degree, a doctoral degree in art or a foreign degree that is deemed to be equivalent to a doctoral degree. This eligibility requirement must be met before the employment decision is made.

In the first instance, those who have completed their degree no more than three years prior to the end of the application period shall be considered. Those who have completed their degree more than three years prior to the end of the application period may also be considered in the first instance if special grounds exist. Special grounds relate to leave of absence due to illness, parental leave, commissions of trust within union organisations, service within the defence services or other similar circumstances, as well as clinical service or service/assignment relevant to the subject area.

Assessment

Previous theoretical knowledge in the field is appreciated. Previous experience of behavioral models in rats and mice as well of the in vivo microdialysis technique and neurochemistry is a merit. Great emphasis will be placed on personal suitability.

  • The Department of Medical Biochemistry and Cell Biology
  • The Department of Laboratory Medicine

At present, the institute has about 360 employees and approximately 450 million SEK in total assets

We are currently seeking several highly talented, innovative and results-driven persons for postdoctoral positions in structure and function of human membrane proteins in the Johansson laboratory at the Department of Medical Biochemistry and Cell Biology, Institute of Biomedicine at the Göteborg University, Sweden.

Subject area

Structure and function of human membrane proteins

Subject area description

G protein-coupled receptors (GPCRs) and ion channels make up two of the largest sub-families of human membrane proteins. These proteins are important targets for therapeutic development and in particular GPCRs since they constitute about 30-40% of all therapeutic drug targets. Methodological breakthroughs have accelerated structure determination of human membrane proteins, however our understanding of the molecular bases of ligand binding and mechanisms of activation remain elusive.  We will characterize these proteins using structural, biophysical and pharmacological methods.

Duties

The successful candidates will work on the structure and function of select human membrane protein targets, including G protein-coupled receptors and ion channels, using a combination of structural, biochemical and pharmacological approaches. The candidates will be encouraged and expected to conduct their research independently and to plan and execute research tasks in a timely manner. The candidates are also expected to travel for data collection purposes and to participate in collaborations with other research groups (either within Sweden or abroad).
